The weekly service will operate from Manchester and Gatwick airports during the summer months, beginning on May 5th and ending on October 5th 2010.
According to the company, the route will operate every Tuesday from both UK airports and can be booked online via its website.
Return flights begin at £160, including all tax and surcharges.
Monarch states: "Formed by an ancient volcanic eruption, Santorini is famed for having the best sunsets in the world and boasts an astonishing blend of scenery, radiant white villages and spellbinding archaeological sites."
It adds that it could be the "dream Greek getaway" for couples and families alike.
The carrier operates flights from a number of other UK airports, such as London Luton and Birmingham, to countries such as Turkey, Portugal, Cyprus and Gibraltar.
It recently announced it will operate additional flights between London Gatwick and Tenerife next month to cope with increased demand.
